The large protein clumps in dairy effluent make it harder for microbes to break down the waste, making traditional biological treatment methods less effective. This research generated an extracellular protease from Pseudomonas aeruginosa, partially purified it, and utilized it as an enzymatic pretreatment to improve substrate accessibility for biological processes. The purification method, which included ammonium sulfate precipitation and dialysis, raised the specific activity from 1.21 to 20.26 U mg −1 . This meant that the active enzyme fraction had been made stronger. Size-exclusion high-performance liquid chromatography (SEC-HPLC) was used to evaluate molecular changes in protein-rich wastewater. Following enzymatic treatment, chromatographic profiles showed a big change from early-eluting high-molecular-weight protein fractions to late-eluting low-molecular-weight peptides. This was clear proof of proteolytic cleavage. This structural transformation significantly altered on the physical and chemical properties of organic nitrogen, but it didn’t change the amount of organic matter. Biodegradability studies showed that samples that had been treated with enzymes used more oxygen in the first stage of decomposition. The high biochemical oxygen demand (BOD) and chemical oxygen demand (COD) levels did not mean that pollution was getting worse instead, they meant that microbial oxidation was happening faster because bacteria could get to soluble substrates more easily. The BOD/COD ratio showed that the speed of biodegradation was getting better. Enzymatic hydrolysis turned slowly biodegradable particulate COD into soluble parts that break down quickly. A mechanistic link exists between protein fragmentation and improved biological treatment effectiveness when enzyme purification, molecular-scale chromatographic analysis, and biodegradation performance evaluation are utilized simultaneously. The findings demonstrate that protease-based pretreatment is a viable and energy-efficient approach for mitigating hydrolysis issues in activated sludge systems. This could be a good way to deal with dairy waste that is high in protein.
This study is designed to isolate and molecular identification of C. neoformans, C. neoformans is pathogenic yeast and effect immunocompromised and immunocompetent. Methods: collect 50 samples from pigeon dropping and 50 samples from pigeon fanciers (sputum). The collection time was extended from November 2021 to February 2022, then culture at SDA, BSA, Cryptococcus Differential agar, esculin agar, Eucalyptus leaves agar media and Brain heart infusion agar with methyldopa, biochemical test including urease test and methyldopa, and then confirm identification by molecular identification by PCR technique sequencing and genetic analysis. The results showed that 3 swaps taken from sputum of human included cryptococcus neoformans and 6 s

Each control cultivar and their derived lines were independently cultured in plastic bags according to the RCBD, with three replications. The results showed that the highest plant height and number of leaves were obtained from Arizona derived lines which gave 60.11 cm and 25.30 leaves.plant-1 in

This range of wavelengths (300–356) nm was chosen. The method showed linearity in the 2-22 μg/mL range, with R2 being 0.9996. The developed method' linearity, detection limit, quantification limit, precision, repeatability, and accuracy were all statistically and experimentally validated. The suggested methodology can be used for routine quality control analysis of Rifampicin in pure form and in capsule dosage form, as demonstrated by the satisfactory recovery percentage results. This study explores the struct

Over time, other forms of treatment have been developed and modified, including extended aeration. This process is included in the suspended growth system. In this paper, a comparative study was conducted between the efficiency of the extended aeration plant and that of the trickling filter plant in removal of BOD and COD. The method of comparison was done by knowing the value of the pollutant before and after the treatment and then extract the removal ratio of each pollutant within each plant. The results showed that the percentage of removal of BOD in the trickling filte

This requires using a sustainable technique to treat natural municipal wastewater as an alternative to conventional methods, especially in arid and semi-arid rural areas. One of the promising techniques applied to satisfy the objective of wastewater reuse is the constructed wetlands (CWs) which have been used extensively in most countries worldwide through the last decades. The present study introduces a significant review of the definition, classification, and components of CWs, identifying the mechanisms controlling the removal process within such units. The chromatographic separations were achieved with a Waters Symmetry® C18 column (5 μm, 4.6 × 150 mm), using a mixture of methanol and water (40:60, v/v) as a mobile phase, under isocratic elution mode with a flow rate of 0.8 mL/min, and ultraviolet (UV) detection was set at 264 nm. The oven temperature for the column was set and maintained at 35 °C. These infections appear to be rising particularly among pets and their owners, suggesting a possible bidirectional transmission between humans and dogs. Therefore, this study aimed to investigate the potential link of mixed infections involving Klebsiella pneumoniae and Enterococcus spp. Buccal cavity samples were collected from humans (n = 25) and dogs (n = 25). Samples were initially enriched in tryptic soy broth and subsequently cultured on tryptic soy agar, MacConkey agar, and blood agar. All isolates were identified using the VITEK 2 system, and eight selec
