The large protein clumps in dairy effluent make it harder for microbes to break down the waste, making traditional biological treatment methods less effective. This research generated an extracellular protease from Pseudomonas aeruginosa, partially purified it, and utilized it as an enzymatic pretreatment to improve substrate accessibility for biological processes. The purification method, which included ammonium sulfate precipitation and dialysis, raised the specific activity from 1.21 to 20.26 U mg −1 . This meant that the active enzyme fraction had been made stronger. Size-exclusion high-performance liquid chromatography (SEC-HPLC) was used to evaluate molecular changes in protein-rich wastewater. Following enzymatic treatment, chromatographic profiles showed a big change from early-eluting high-molecular-weight protein fractions to late-eluting low-molecular-weight peptides. This was clear proof of proteolytic cleavage. This structural transformation significantly altered on the physical and chemical properties of organic nitrogen, but it didn’t change the amount of organic matter. Biodegradability studies showed that samples that had been treated with enzymes used more oxygen in the first stage of decomposition. The high biochemical oxygen demand (BOD) and chemical oxygen demand (COD) levels did not mean that pollution was getting worse instead, they meant that microbial oxidation was happening faster because bacteria could get to soluble substrates more easily. The BOD/COD ratio showed that the speed of biodegradation was getting better. Enzymatic hydrolysis turned slowly biodegradable particulate COD into soluble parts that break down quickly. A mechanistic link exists between protein fragmentation and improved biological treatment effectiveness when enzyme purification, molecular-scale chromatographic analysis, and biodegradation performance evaluation are utilized simultaneously. The findings demonstrate that protease-based pretreatment is a viable and energy-efficient approach for mitigating hydrolysis issues in activated sludge systems. This could be a good way to deal with dairy waste that is high in protein.
High performance liquid chromatography was applied for the separation and identification of four antharquinone derivatives, aloe emodin, emodin, chrysophanol and physcion. The separation was carried out using Eurospher 100, C18 column (4.6 mm i.d. x 250 mm, 5 µm) under the following conditions: acetonitile (solvent A) and water: acetic acid (99.9: 0.1 v/v, pH 3.5)( solvent B) as a mobile phase with isocratic elution with 30% solvent B at flow rate 0.8 ml/min. The detection wavelength was set at 254 nm. The four antharquinone derivatives were isolated from the Iraqi rhubarb, Rheum ribes root by preparative TLC, their structures were identified by 1H NMR and used as standards for HPLC analysis. The percentages of alo

STa activity was estimated by using suckling mouse assay (SMA) and from these (11) isolates only (5) showed STa activity and the one with the highest STa activity was selected for large scale production of STa, which was followed by partial purification using ion-exchange chromatography (normal phase) using DEAE sephadex A-50 column. The family Acetobacteraceae includes the obligate aerobic, motile acetic acid bacteria. The BNC has attracted a lot of interest across a wide range of industries, including pharmaceuticals, due to its flexible characteristics, properties, and advantages. The present study was conducted to purify and characterize BNC produced from AAB isolated from apple vinegar. Bacterial nanocellulose was synthesized using a natural date palm liquid medium at pH 6 at 30°C for 8–10 days. The bacterial cellulose produced was then purified using a technique involving 0.1 M sodium hydroxide. To ascertain the surface mor
