From adhesion to gene regulation: tea tree essential oil suppresses Uropathogenic Escherichia coli colonization while triggering csgA-mediated biofilm stress paradox

Background and objectives Uropathogenic E.coli (UPEC) are major causative agents of urinary tract infection (UTIs), they often possess strong biofilm-forming abilities, and capable of resisting many antibiotics, making catheter associated UTIs (CAUTIs) difficult to treat. Essential oils such as tea tree oil (TTO) have emerged as natural alternatives to antibiotics. This study aimed to evaluate the antibacterial, adhesion, and biofilm-forming efficacy of TTO against UPEC, while analyzing its effect on the gene expression of csgA gene and determining its cytotoxicity. Methods Four UPEC isolates collected from different UTIs patients from Baghdad Province. The Antibacterial activity of TTO evaluated using agar wells diffusion assay and micro dilution using resazurin. Anti-adhesion and anti-biofilm were assessed using silicon Foley catheters. The csgA encode to curli fibers determined using polymerase chain reaction (PCR) and gene expression measured using qPCR. Cytotoxicity of TTO measured against renal carcinoma (A498) and normal fibroblast (HdFn) cell lines via MTT assay. Results TTO inhibited UPEC with inhibition zone diameter of 12–25 mm (p < 0.0001) and MIC value was 0.25%. In Foley catheter model, the concentrated TTO reduced adhesion and biofilm formation (p < 0.0001). csgA harbored within all subjected isolates. Real time quantitative PCR (RT-qPCR) revealed significant (p < 0.0001) upregulation within susceptible isolates (2.2) fold change. Cytotoxicity via MTT assay reveled selective activity of TTO on (A498, IC50= 265.8 µg/mL) over (HdFn, IC50 = 852.5 µg/mL; p < 0.0001). Conclusion TTO demonstrated potential antibacterial, anti-adhesion and anti-biofilm activity against UPEC along with modulations of csgA gene expression and selective cytotoxicity

A total of (90) blood samples were collected from male patients infected with Toxoplasmosis who recovered from COVID- 19 and attended Kamal Alsamiraai Hospital from 15 January to 15 September 2021. We measured anti-Toxoplasma antibodies (IgG and IgM) detected by ELISA, whereas Anti-COVID-19 antibodies (IgG and IgM) were estimated using Elisa and Afilias. Mutations in ergosterol 11 gene of fluconazol resistant candida albicans isolated from different clinical samples

Fluconazole was used to test the susceptibility of Candida albicans isolated from different clinical samples, and to detect mutations in ERG11 gene, and their relationship to fluconazole resistance. Forty-eight isolates of Candida albicans were tested for susceptibility using the disc diffusion method (M-44). ERG11 genes of six isolates were amplified (four resistant, two susceptible) and sequenced. The sequenced genes were analyzed to detect the mutations. Extraction of Oil from Eucalyptus Camadulensis Using Water Distillation Method

This work was conducted to study the extraction of eucalyptus oil from natural plants (Eucalyptus camaldulensis leaves) using water distillation method by Clevenger apparatus. The effects of main operating parameters were studied: time to reach equilibrium, temperature (70 to100°C), solvent to solid ratio (4:1 to 8:1 (v/w)), agitation speed (0 to 900 rpm), and particle size (0.5 to 2.5 cm) of the fresh leaves, to find the best processing conditions for achieving maximum oil yield. The results showed that the agitation speed of 900 rpm, temperature 100° C, with solvent to solid ratio 5:1 (v/w) of particle size 0.5 cm for 160 minute give the highest percentage of oil (46.25 wt.%). The extracted oil was examined by HPLC.

Kinetics and Thermodynamics of Peppermint Oil Extraction from Peppermint Leaves

This study aimed to extraction of essential oil from peppermint leaves by using hydro distillation methods. In the peppermint oil extraction with hydro distillation method is studied the effect of the extraction temperature to the yield of peppermint oil. Besides it also studied the kinetics during the extraction process. Then, 2nd -order mechanism was adopted in the model of hydro distillation for estimation many parameters such as the initial extraction rate, capacity of extraction and the constant rat of extraction with various temperature. The same model was also used to estimate the activation energy. The results showed a spontaneous process, since the  Gibbs free energy had a value negative sign.
