Effect of dietary parsley (Petroselinum crispum) supplementation on semen quality of local Iraqi ganders

This study was carried out to investigate the effect of dietary supplementation with different levels of parsley on semen quality of local Iraqi ganders. A total of thirty-two local ganders were used in this study during the period from the beginning of February to the end of April. The ganders were allocated for 4 treatment groups containing 8 ganders each. The treatment groups were as follows: Control diet (free from parsley); T1: Control diet + 80 g/d parsley; T2: Control diet + 160 g/d parsley; T3: Control diet + 240 g/d parsley. Semen samples were collected twice a week, fortnightly, from each gander by the dorsal-abdominal message method. The first semen collection was used to evaluate semen volume, sperm concentration, live in total sperm, live and normal morphology sperm, semen quality factor, sperm motility, abnormal sperm, acrosomal abnormalities, spermatocrit, and pH of semen However, the second semen collection was used to determine seminal plasma concentrations of glucose, protein, cholesterol, and other activities of aspartate aminotransferase (AST), alanine aminotransferase (ALT), and alkaline phosphatase (ALP) enzymes. Results revealed that feeding diets containing different levels of parsley (T1, T2, and T3) resulted in a significant (P<0.05) increase in semen volume, sperm concentration, live and normal morphology sperm, semen quality factor, sperm motility, spermatocrit, and seminal plasma activity of the ALP enzyme, and a significant (P< 0.05) decrease in abnormal sperm and acrosomal abnormalities, seminal plasma concentrations of glucose, protein, and cholesterol, and seminal plasma activities of the AST and ALT enzymes as compared with the control group. There was no significant difference in the pH of semen among the control and experimental groups (C, T1, T2, and T3). Additionally, adding different amounts of parsley to the diet, especially 240 g/d (T3), led to big improvements in the characteristics of the sperm. So, parsley can be used as an effective tool to improve the semen quality of ganders.

Purification and Characterization of Endoglucanase from local isolate of Aspergillus flavus

Endoglucanase produced from Aspergillus flavus was purified by several steps including precipitation with 25 % ammonium sulphate followed by Ion –exchange chromatography, the obtained specific activity was 377.35 U/ mg protein, with a yield of 51.32 % .This step was followed by gel filtration chromatography (Sepharose -6B), when a value of specific activity was 400 U/ mg protein, with a yield of 48 %. Certain properties of this purified enzyme were investigated, the optimum pH of activity was 7 and the pH of its stability was 4.5, while the temperature stability was 40 °C for 60 min. The enzyme retained 100% of its original activity after incubation at 40 °C for 60 min; the optimum temperature for enzyme activity was 40 °C.

Separation and Purification of Hemolysin from Local Isolate of Serratia marcescens

Twenty isolates of Serratia marcescens were isolated from inflammation of the urinary tract (UTI)., These isolates were found to produce hemolysin as indicated by blood agar plates in which the hemolysis of red blood cell indicate a positive result. Isolates were selected according to their hemolysis activity by measuring absorbance of hemoglobin at 405 nm that released from red blood cell. Hemolysin was completely purified using 50-75% saturation of ammonium sulphate followed by ion exchange chromatography with DEAE-cellulose then gel filtration chromatography by sepharose 4B. Accordingly molecular weight for the purified toxin was estimated as 45 KD.

Determination of chemical composition of local black seeds Nigella sativa L

Chemical analysis for evaluation of Nigella sativa L. (black cumin) seeds showed a composition of Fat 39% ; Protein 28% ; Carbohydrate 21% ; Moisture 6% and Ash 4.5% . It was found that the black seed contains the following mineral element : Magnesium 0.26 gm /100gm seed ; Calcium 0.25 gm /100gm seed and Iron 25 ?g / gm /100gm seed ; zinc 4.51?g /gm /100gm seed and Copper 3.60 ?g /gm /100gm seed. The analysis also showed that mineral element I. e. ; lead ; Cobalt ; Nickel ; Chrom ; Cadmium and Aresenic are not present . It was found that the fat of the black seed contains the following fatty acids : Myristic 2.8%; Palmtic 16.6%; Stearic 0.8 % ; Oleic 13.79% ; Linoleic 64.2% and Arachidic 1.9% .
