This study was aimed to isolate and identify Saccharomyces boulardii from Mangosteen fruits (Garcinia mangostana L.) by traditional and molecular identification methods To get safe and healthy foods probiotics for use, The isolates and two commercial strains were subjected to cultural, morphological and biochemical tests, The colonies of the isolates were spherical, smooth, mucoidal, dull and white to cream colour on SD agar media .The shape of cells was globose to ovoid and sometimes with budding, in a single form or clustered like a beehive. The isolates and two commercial strains were unable to metabolized galactose and lactose , Results shows that all isolates were unable to utilize potassium nitrate and not grow in the presence of (0.01%) cyclohexamide. Also the isolates and two commercial strains were identified by the Vitek 2 identification system, as S. cerevisiae with probability 90-94%, Since there are no data in this device includes S. boulardii. Because the cultural, morphological and biochemical tests didn’t provide sufficient evidence to distinguish between S. boulardii and S. cerevisiae, the probiotics strains must be identified up to genus and strain levels by internationally accepted methods, So the SbR7 isolate which shown high probiotic advantages and two commercial strains were diagnosed molecularly by using specific primers targeting sequence for the region (ITS) of the 5.8S rRNA gene Genomic DNA was isolated from SbR7 Isolate and ITS region of the 5.8S rRNA gene was amplified using PCR. PCR products was sequenced and compared with the sequence of this region in the DNA of S. boulardii available in GenBank (NCBI) using the program BLASTn. Results revealed, this isolate was almost genetically identical (99%) with S. boulardii standard strains.
تم التحري عن انتاج انزيم الكايتنيز من خميرة Saccharomyces cerevisiae حيث زرعت هذه الخميره في وسط سائل يحتوي على الكايتين ومواد نتروجينية مختلفة . اجريت غربلة لسلالات الخميرة المستعملة لتعين السلالة الاعلى انتاجا للانزيم . بينت النتائج ان السلالة S2 اعطت اعلى فعالية نوعية بلغت 3.5 وحدة /ملغم بروتين .
Abstract
The catalytic cracking conversion of Iraqi vacuum gas oil was studied on large and medium pore size (HY, HX, ZSM-22 and ZSM-11) of zeolite catalysts. These catalysts were prepared locally and used in the present work. The catalytic conversion performed on a continuous fixed-bed laboratory reaction unit. Experiments were performed in the temperature range of 673 to 823K, pressure range of 3 to 15bar, and LHSV range of 0.5-3h-1. The results show that the catalytic conversion of vacuum gas oil increases with increase in reaction temperature and decreases with increase in LHSV. The removal of water turbidity by using crumb rubber filter was investigated .The present study was conducted to evaluate the effect of variation of influent water turbidity (10, 25 and 50 NTU), media size (0.6and 1.14mm), filtration rate (25, 45 and 65 l/hr) and bed depth (30 and 60 cm) on the performance of mono crumb rubber filter in response to the effluent filtered water turbidity and head loss development, and compare it with that of conventional sand filter.Results revealed that 25 l/hr flow rate and 25 NTU influent turbidity were the best operating conditions. smaller media size and higher bed depth gave the best removal efficiency while higher media size and small bed depth gave lower head

In this study, the stress-strength model R = P(Y < X < Z) is discussed as an important parts of reliability system by assuming that the random variables follow Invers Rayleigh Distribution. Some traditional estimation methods are used to estimate the parameters namely; Maximum Likelihood, Moment method, and Uniformly Minimum Variance Unbiased estimator and Shrinkage estimator using three types of shrinkage weight factors. As well as, Monte Carlo simulation are used to compare the estimation methods based on mean squared error criteria.
