The effect of surlactin produced by Lactobacillus acidophilus on eye infectious bacteria in rabbits

Twenty five vaginal swabs from outpatients' healthy women were collected from Kamal Al-Samarai Hospital, Baghdad, to isolate and identify of Lactobacillus acidophilus. Three isolates were diagnosed as L. acidophilus which represents 15% of the total number of lactic acid bacterial (LAB) isolates; other LAB types represent 65% (20 isolates).The ability of L. acidophilus to produce surlactin was detected after measuring its biological activity to inhibit the adhesion of biofilm formed by Pseudomonas aeruginosa to surfaces using test tube method. It was found that all isolates were able to produce surlactin but the activity of surlactin was varying in each isolate. Surlactin produced by isolates 1 and 13 was the most effective. Biological applications of surlactin were studied by inhibiting the adhesion of pathogenic P. aeruginosa producing biofilm on contact lenses. In this study the surlactin has the ability to inhibit the adhesion up to 60% and 55% for isolates 1 and 13 respectively and does not have an antibacterial activity.Surlactin showed an ability to treat the infection in rabbits' eyes with P. aeruginosa while it did not show this ability against Staphylococcus aureus. Additionally, it prevented the infection with P. aeruginosa when administrated to rabbits' eyes inoculated with these bacteria only, while it showed no effect against S. aureus.

Recovery of pure Hesperidin from Iraqi Sweet Oranges Peel and study the effect in some bacteria

Citrus fruit contain variety of flavonoids such as Hesperidin (the principal flavonoid in oranges and grapefruit). Hesperidin is found in high concentration in fruit peel of oranges and in substantially lower concentration in juice of these fruits. Hesperidin was extracted from oranges peel by treating the peels with calcium hydroxide. HPLC technique was used to determine hesperidin. Hesperidin was saperated and purified in a purity of about 90.1-95.7% and yield about 1.5 %w/w from oranges peel dry powder. This research is motivated by the heightened sensitivity of rabbits to infestations by hard ticks, which are recognized as the primary vectors of certain blood-borne diseases, particularly Viral Hemorrhagic Fever (VHF). Antibacterial Activity of Lactobacillus buchneri Bacteriocin against Vibrio parahaemolyticus

Eleven yoghurt samples were collected from local markets in Baghdad to isolate Lactobacillus buchneri. Only 3 isolates of L. buchneri were found and the isolate No. 3 was the most producer of bacteriocin. Bacteriocin was adsorbed 100% onto silicic acid at pH 6.0-7.0. Below or above these pH values, adsorption was decreased, ranging between 35 and 90%. Therefore, pH 6.0 was used for the purification procedure. The purification procedure including silicic acid adsorption/desorption and cation-exchange chromatography (CEC) resulted in a 11.11 fold increase in the final specific activity of pure bacteriocin (1176.47 Au/mg) compared to the culture supernatant which was 32.64 Au/mg.
