UPLC-ESI-MS/MS and Various Chromatographic Technique for Identification of Phytochemicals in Populus euphratica Oliv. Leaves Extract

The aim of this study is to screen the phytochemicals found in Populus euphratica leaves since this type of trees are used traditionally by many villagers as treatment for eczema and other skin disease and also this plant is poorly investigated for their phytochemicals especially in Iraq. Phytochemical screening of the extracts obtained from the n-hexane and chloroform fraction of leaves of Populus euphratica was done by Thin-layer chromatography and various spraying reagents to test if alkaloids, sterols and other compounds are present. UPLC-electrospray ionization –tandem mass spectroscopy along with GC-MS and HPTLC are used to identify the phytochemicals present in the plant leaves.UPLC-ESI-MS/MS method 20 compounds have been identified in various fractions among which are protopine alkaloids. salicin, salicortin, tremulacin. GC-MS showed that the observed data obtained are matched with that in NIST library and confirmed the presence of Hexadecanoic acid trimethylsilyl ester in 43.80% beta-Sitosterol in 37.14% and Diisooctyl phthalate 11.46%.UPLC-ESI-MS/MS is a powerful method for the identification of compounds in mixture based on comparison of their molecular, weight retention time and MS/MS fragmentation. Protopine alkaloid is identified for the first time in Populus euphratica and genus Populus. GC-MS is a valuable method for both qualification and quantification of various phytochemicals that are volatile in nature

Effecienty of Immobilization technique of Bacillus Sublilis Asperginase producer

Abstract Twenty Bacillus isolated were obtained from different sample food and water. Bacillus B1 isolated was the highest asparaginase producer, it was identified as a strain of B. subtilis. The highest production of asparaginase was observed when mineral salt medium containing 0.3% asparagen, pH 8 and incubated at 40°c for 24 hrs. B. subtilis B1 cells were immobilized by entrapment methods (calcium alginate and agar), and by adsorption on solid surface such as sawdust and cotton. The result showed that the immobilized cells by adsorption on sawdust was the best, the immobilized cell retained 88% of asparginase activity after 48h while free cell retained 65%. The aqueous extract of Citrullius colocynthis dried seeds (160 ?g/ml) was in vitro evaluated for its effect on phagocytic index (PI) and lymphocyte transformation index (LTI) of blood cells obtained from 30 apparently healthy blood donors (15 males and 15 females). The PI was further in vivo evaluated in cells of peritone, spleen and liver of mice treated with the extract at a dose of 0.64 mg/kg. The results revealed that in in vitro study, phagocytic cells treated with the extract showed a significant increased percentage as compared with untreated cells (60.0 vs. 44.1%). In this study, we aimed to investigate the potential wound-healing activity of the methanol extract derived from Allium ampeloprasum. Using 30 healthy male Wester rats; control group: 15 rats; normal saline applied locally instead of plant-extract. Experimental group: 15 rats received a daily local application of 200 mg/kg of the plant extract. Then, for each healing period of 0, 5, or 10 days, each group was divided into three-subgroups: A, B, and C, each with five rats. In the process of synthesizing silver nanoparticles we detected a rapid reduction of silver ions leading to the formation of stable crystalline silver nanoparticles in the solution.
