Anti-bacterial Properties of Melatonin against Mycobacterium Tuberculosis in Vitro

         57 isolates of Mycobacterium tuberculosis and Mycobacterium bovis were identified; they were isolated from different clinical sources which included sputum, bronchial wash, abscess, pleural fluid, gastric fluid, eye fluid, and CSF, also urine and ear swab. This investigation was carried out on 198 patient attended National Reference Laboratory for T.B during September 2009. Also the study declared that the ratio of separation of this bacterium from male was (67.6%) and it’s higher than the ratio of separation this bacterium from females which was (32.3%). The susceptibility of Mycobacterium tuberculosis to melatonin was evaluated. Many concentrations of melatonin were prepared to investigate it as antibacterial drug against multidrug resistant Mycobacterium tuberculosis and Mycobacterium bovis. Suspension bacteria (10-1, 10-3 and 10-5) were cultured on Lowenstein-Jensen media (LJ) contains melatonin, while control media without this drug. Six isolates were chosen according to their susceptibility patterns; they were resisting to Rifampicin, Streptomycin, Isonicotinic –hydrazide and sensitive to Ethambutol. In conclusion, these in vitro studies clearly demonstrate anti-bacterial effects of melatonin. Among possible mechanisms, it is concluded that melatonin showed antibacterial effects against multidrug resistant T.B by reducing intracellular substrates. Identifying the mode of action could be of great help in developing and researching new anti-bacterial drugs.

Developing of Bacterial Mutagenic Assay System for Detection of Environmental and Food MutagensV – Using Anticancer Drug Cyclophosphamide

G-system composed of three isolates G3 ( Bacillus),G12 ( Arthrobacter )and G27 ( Brevibacterium) was used to detect the mutagenicity of the anticancer drug, cyclophosphamide (CP) under conditions similar to that used for standard mutagen, Nitrosoguanidine (NTG). The CP effected the survival fraction of isolates after treatment for 15 mins using gradual increasing concentrations, but at less extent comparing to NTG. The mutagenic effect of CP was at higher level than that of NTG when using streptomycin as a genetic marker, but the situation was reversed when using rifampicin resistant as a report marker. The Antibiofilm Efficacy of Gold Nanoparticles Against Acinetobacter baumannii

Acinetobacter baumannii is highly adapted to hospital environments, causing persistent chronic infections due to its ability to form biofilms. In this work, the antibiofilm activity of AuNPs with a subMIC concentration of 9.34 μg/ml was investigated by the microtiter plate method against 80 clinical isolates of A. baumannii. The results revealed that the biofilm was significantly (P< 0.05) reduced by 48.2 – 82.1%.

Antibacterial Activity of Lactobacillus buchneri Bacteriocin against Vibrio parahaemolyticus

Eleven yoghurt samples were collected from local markets in Baghdad to isolate Lactobacillus buchneri. Only 3 isolates of L. buchneri were found and the isolate No. 3 was the most producer of bacteriocin. Bacteriocin was adsorbed 100% onto silicic acid at pH 6.0-7.0. Below or above these pH values, adsorption was decreased, ranging between 35 and 90%. Therefore, pH 6.0 was used for the purification procedure. The purification procedure including silicic acid adsorption/desorption and cation-exchange chromatography (CEC) resulted in a 11.11 fold increase in the final specific activity of pure bacteriocin (1176.47 Au/mg) compared to the culture supernatant which was 32.64 Au/mg.
