Molecular Study of Quercetin-Resistant Pseudomonas aeruginosa Isolated from Wound Infections
...Show More Authors
Abstract
<title>Background:

Quercetin, a crucial plant flavonoid, exhibits a range of pharmacological properties. A significant body of literature explores the antibacterial activity of the substance and its potential mode of action. Quercetin has shown the capacity to inhibit the growth of several types of gram-positive and gram-negative bacteria, as well as fungi and viruses.

Objectives:

The work presented sought to identify siderophore genes in quercetin-resistant Pseudomonas aeruginosa. Bacterial isolates from swab samples taken from patients suffering from complicated wound infections.

Materials and Methods:

A total of 200 swab samples were collected from attending patients. Compared to patients who are male (41%), the majority of patients (59%) were female. The patients have provided swab samples for sterile swab transport, displaying signs of a wound infection. The samples were cultivated on nutrient agar, blood agar, MacConkey agar, and cetrimide agar, and they were incubated for 24 h at 37°C.

Results:

The results were recorded by isolating gram-negative bacteria and observing developed colonies after identifying n = 59 (29.5%) isolates from 200 clinical samples as P. aeruginosa . The plcN and exoS genes were found in the isolates chosen. In this study, n = 10 (100%) isolates tested positive for the 16s rRNA housekeeping gene, n = 8 (80%) of isolates tested positive for the plcN gene, and n = 8 (80%) isolates tested positive for the exoS gene.

Conclusion:

The high prevalence of plcN and exoS gene detection in quercetin-resistant strains may be the primary reason for the high virulence factor in connection with quercetin resistance.

ANTIBACTERIAL AND ANTIBIOFILM EFFECTS OF BISMUTH NANOPARTICLES PRODUCED BY BACILLUS SUBTILIS AGAINST MULTIDRUG-RESISTANT Pseudomonas aeruginosa
...Show More Authors

This study aimed to determine the effect of green bismuth oxide (BiO) NPs against multidrug-resistant (MDR) Pseudomonas aeruginosa (P. aeruginosa) from wound infections. Among 450 wound samples collected from patients admitted to the hospital, 200 P. aeruginosa isolates were identified. MDR strains of P. aeruginosa were detected by disc diffusion method. BiO NPs were synthesized using wild Bacillus subtilis (B. subtilis) strain and infrared spectroscopy, X-ray diffraction and scanning electron microscopy techniques. The antibacterial effect of the NPs compared to antibiotics against MDR strains was evaluated using a standard disk diffusion method. BiO NPs were synthesized at 0.005 M concentration of solution. According to the SEM im

Incidence of Ciprofloxacin-Resistant of Methicillin Resistant Staphylococcus aureus isolated from Iraqi patients
...Show More Authors

The resistance of Staphylococcus aureus to ciprofloxacin has complicated the problem of treating staphylococcal associated infections in which MRSA is the causative agent since ciprofloxacin was the drug of choice to treat such infections. Our study investigated the incidence of Ciprofloxacin resistant S. aureus isolates that were also methicillin resistant among Iraqi patients. The obtained bacterial isolates were tested for Ciprofloxacin resistance using agar dilution method and the sequence of gyrA and parC. The results revealed that about 8% of the isolated MRSA strains were Ciprofloxacin resistant and the resistance was due to mutation in gyrA rather than parC.

Study of Some Physical and Chemical Properties of Staphylolysin Enzyme purified from Pseudomonas aeruginosa
...Show More Authors

Some of the characters of the Staphylolysin A and D enzymes purified from Pseudomonas aeruginosa P16 and P5 respectively were studied, the molecular weights of Staphylolysin A and D were 20.417 kilo dalton and 23.988 kilo Dalton respectively by SDS- polyacryl amide gel electrophoresis. The optimum pH for staphylolysin A activity was found to be 8 which gives higher activity reaches 150 unit/ml, and for enzyme stability was 7.5-8.5 in which the enzyme nearly retained its full activity, while it was 9.5 for staphylolysin D that gives higher activity of 16 unit/ml,and 8.5-9.5 for enzyme stability in which the enzyme nearly retained its full activity, Maximum activity of two enzymes was obtained at 40C in which the specific activity for st

Comparative Study of Some Antibiotic Resistance, Extended Spectrum ß-Lactamases (ESBLs), Metallo ß-Lactamases (MBLs) Produced in Pseudomonas aeruginosa Isolated from Clinical and Food Samples
...Show More Authors

A total number of 33 isolates of Pseudomoans aeruginosa were collected from different clinical samples, such as: burn, wound and urine from patients attending Al-Yarmouk teaching hospital and some private clinical laboratories in Baghdad city through the period from October to December 2016. On the other hand, 21 isolates of P. aeruginosa were collected from 38 different food samples; such as: vegetables and fruits, from different local markets in Baghdad city during the period from November to December 2016. All isolates were identified by using different bacteriological and biochemical assays and confirmed by Vitek-2 identification system. The antimicrobial susceptibility test for clinical and food isolates towards 17 antimicrobial a

Tigecycline is the Most Effective against Multi-Drug Resistant Klebsiella pneumoniae Recovered from Burn Wound Infections in Two Hospitals in Al-Kut City, Iraq
...Show More Authors

Klebsiella pneumoniae is among the most frequent microorganisms isolated from infections of burn wounds. This cross-sectional study aimed to investigate the distribution of multi-drug resistant (MDR) K. pneumoniae in two burn hospitals and the antibiotic resistance profile in different burn regions of the same patient. It was performed in two hospitals (Al-Zahraa and Al-Karama) in Al-Kut, Iraq, between January and May 2022. Totally, 100 burn swabs were collected from 40 patients of both genders suffering from burn wound infections, with ages ranging between 3 and 50 years. Klebsiella pneumoniae were isolated and identified using conventional methods followed by VITEK®2 system and confirmed via polymerase chain reaction targeting t

Studying Proteus mirabilis Typing Isolated from Urinary Tract infections
...Show More Authors

Urine specimen (253 samples) had been collected from urinary tract infections. The study showed that Proteus mirabilis was responsible for (11.85%) of the urinary tract infections. Also, the study had declared that the ratio of isolation of this bacterium from women was (7.51%) and it was higher than the ratio in both men and children which ranged (1.58%) and (2.76%) respectively. Morphological and biochemical studies had been applied to characterize this bacterium as well as other kinds of microorganisms that were isolated from urinary tract infection in this study. Evaluation of Synergistic Antibacterial Effect of Combined Scrophularia striata Extract and Antibiotics Against Pseudomonas aeruginosa and Methicillin -Resistant Staphylococcus aureus

Scrophularia. striata from Scrophulariacea family has been used in Iranian folk medicine for the treatment of infectious diseases. In this study we evaluated the synergistic effect of S. striata   hydroalcoholic extract (SSE) and commercially available antibiotics against P. aeroginosa and Methicillin- resistant Staphylococcus aureus (MRSA). The resazurin-based microdilution method was used to determine the minimum inhibitory concentration (MIC) values of plan extract and standard antibiotics. The interaction between standard antibiotics and SSE was evaluated by using checkerboard method. The results of this study revealed that SSE enhance the antibacterial activity of antibiotics. The combin
