Effect of Volatile oil extracted from Citrus grandis yellow peel on growth of some food borne microorganisms

     The study is conducted to investigate the effect of volatile oil extracted from the yellow peel of C. grandis fruits (Sindi) on growth of three bacterial species isolated from food samples , as well as A. flavus , and to check the possibility of using the volatile oils of this plant as a food preservative .     Results showed a variation in the sensitivity of the bacterial isolates against the volatile oil which showed inhibitory effect on the growth of S. enteritidis and    S. aureus with the increasing concentration of the volatile oils used in this studys , MIC for both bacteria was (0.12 , 1)% (v:v) respectively. Whereas , P. aeruginosa showed complete resistance to all treating concentrations used . Results of treating spore suspension of A. flavus with different concentrations of volatile oil showed also complete inhibition of germination at concentrations (0.12 , 0.25 , 0.5 , 1)%. Whereas lower concentrations (0.03 , 0.06)% delayed spore germination for (11) and (17) hrs after incubation respectively compared to the control treatment in which spores germination was within (6) hrs after incubation .      Different concentrations of the volatile oil also affected the radial growth rate of A. flavus was colonies with the increasing concentrations . Preparation of Media for Growth and Isolation of Lactic Acid Bacteria by Using the Starch-Industry by- Products

By- products of corn starch industry were used to prepare media for propagation the lactic acid bacteria as a natural auxotroph. The by- products used were the corn steep water (S) and gluten extract (G) after a proper treatment to get them ready for media preparation. The results showed that it was possible to replace the peptone and meat extract by gluten extract in MRS medium. The growth was approximately similar to that obtained in standard MRS media. This study included the estimation of growth rate, viability and morphological changes in different culture media (NNN, P-Y, RPMI- 1640, and Panmed). Promastigotes cultured in RPMI-1640 showed maximal growth rate after (2, 4, 6) days of cultivation (27.26 ± 0.05), (172.20 ± 0.1) and (343.81 ± 1.48) million parasites / ml for each day respectively, while P-Y media gave the highest mean of growth rat after (8 and 10) days of cultivation (307.16 ± 1.67) and (303.5 ± 4.96) million parasites / ml for each day respectively. P-Y medium showed the maximal percentage of viability after (2, 4, 6, 8, and 10) days of cultivation (99.76 ± 0.5) %, (98.30 ± 0.17) %, (96.1 ± 0.1) %, (92.5 ± 0.52) % and (87.26 ± 0.05) % for each day respectively.

Data collected during the period extended from November, 2002 to October, 2003 except for the month of April The population consisted of five age groups; O, I, II, III, and IV which have 0, 1, 2, 3 and 4 annuli respectively. The study also proved the validity of annuli readings for age and growth determination.
