Background: Breast cancer has become one of the most predominant health risks among women and its mass death rate has continued to escalate world over. New data indicate that breast cancer may be modification by lifestyle factors, especially diet. Lactobacillus and Lactococcus type of probiotics have been identified to cure or promote health by regulation of the immune mechanism. Such bacteria are frequently found in healthy breast tissue and they could possibly help prevent breast cancer. Combining with a well established probiotic, Bifidobacterium longum, Lactobacillus acidophilus has proved to have immunomodulatory and anti-inflammatory effects, therefore, it should be a prospective probiotic strain to maintain breast health. Aim: was to assess the inflammatory effect of the probiotic supplement on selected breast cancer cell lines. Methods: This study examined the response of two distinct probiotic supplements on the immune-related responses within the cell line of breast cancer (AMJ-13 and MCF-7).The first probiotic (T1) consisted of Lactobacillus plantarum 299v, while the second (T2) was a bacterial cocktail comprising eight probiotic strains: Lactobacillus acidophilus, Lactobacillus rhamnosus, Bifidobacterium bifidum, Bifidobacterium breve, Streptococcus thermophilus, Lactobacillus casei, Lactobacillus plantarum, and Lactobacillus acidophilus. Their immune responses were evaluated in terms of IL-22 (a pro-inflammatory cytokine) and IL-37 (an anti-inflammatory cytokine) levels measured so via ELISA assays at 48 and 72 hours. Results: The findings revealed that the T2 probiotic cocktail had strong effects on preventing IL-22 release in both MCF-7 and AMJ-13 cells after the two time points (p < 0.05). Instead, T1 played a significant proliferative role on increasing IL-22 level in AMJ 13 cell at 48 and 72 hours. Moreover, T1 had a great impact on IL-37 production at both timepoints whereas T2 regulated IL-37 secretion both in MCF-7 and in AMJ-13 cells. These results indicate that probiotic bacteria could work on immune responses through regulating the release of inflammatory cytokines, maybe by intervening in the inflammatory milieu linked to breast cancer. Conclusion: Summing up, probiotic supplements, especially, preparations with live bacteria could potentially modulate immune responses and interfere with inflammation in breast cancer. These findings emphasize the significance of the continued study of the probiotic use in the treatment of breast cancer.
Toxoplasmosis is a parasitic infection that triggers immune cells to produce cytokines and inflammatory mediators that are responsible for abnormal or aborted immune responses. This study highlights the evaluation of the Dectin-1 receptor and cytokine IL-37 in the serum of 80 patients who had miscarried in the first trimester and were infected with toxoplasmosis, as well as 40 pregnant women in the first trimester who had a successful pregnancy (control groups). The serum was first screened for the T. gondii IgM and IgG antibodies by an enzyme-linked immunosorbent assay (ELISA) and then the serum levels of IL-37 and Dectin-1 were determined. The results showed that the serum level of Dectin-1 was significantly increased in anti-

Estimation of the in situ expression of IL-6 and IL-1β in breast cancer patients. A sixty patients with breast cancer BC were divided into two clinical subgroups, (30) with malignant breast cancer MBC and (30) with benign breast tumor as a control group according to histological examination. In situ hybridization technique used for detection of IL-6 and IL-1β mRNA sequence in two groups. The results showed that percentages of mRNA expression of IL-6 and IL-1β were in (≥ 11-50%) for malignant breast cancer. This research also investigated that (73.3%) of beni

Specific Background: Recent research has focused on the role of epigenetic changes in DNA damage, repair mechanisms, and the potential therapeutic effects of probiotics. Probiotics have shown promise in promoting tissue regeneration and DNA repair. Knowledge Gap: However, the precise impact of probiotics on DNA repair in cancer cells, specifically breast cancer cells, remains underexplored. Aims: This study aimed to evaluate the effects of probiotics on DNA damage repair in AMJ13 Iraqi breast cancer cells and assess the cytotoxic effects of probiotics on these cells. Results: Using the comet assay, we found significan

In this study we explored the anticancer activity of genetically modified NDV expressing IL-2-P53 (rClone30–IL-2-P53) in lung cancer model. We have cloned IL-2 and P53 genes and inserted them in the viral genome of New Castle Disease Virus to create a genetically modified rNDV- IL-2-P53 virus and tested the anti-tumor activity of the new virus in vitro on different types of cancer cell lines by MTT assay. TheIL-2 and P53 gene were successfully cloned and inserted into the viral genome by using a Mlu I and Sfi I endonucleases, viral vector was constructed correctly and successf

BRCA1 transcript levels were assessed by real time quantitative polymerase chain reaction (RT-qPCR) in the cancer cell lines. Our data show overexpression of BRCA1 mRNA level in all the studied breast cancer cell lines: MCF-7, T47D, MDA-MB-231 and MDA-MB-468 along with Jurkat, leukemia T-lymphocyte, the positive control, relative to normal breast tissue. To investigate whether a positive or negative correlation exists between BRCA1 and the transcription factor E2F6, three different si-RNA specific for E2F6 were used to transfect the normal and cancerous breast cell lines. Interestingly, strong negative relationship was found b

The aim of the study is investigation and detection of some bacterial interfering with CRC occurrence and progression. The study conducted between September 2022 till February 2023, a total of 50 specimens were collected from confirmed CRC patients. In addition, 50 stool specimens were collected from Healthy volunteers, considers as control group. Isolation and identification of bacteria in all collected specimens were done by using cultural and differential media (blood agar, macconkey agar and Pfizer agar), as well as the VITEK- 2 compact system. The bacterial species, in the specimens of control were ( Escherichia coli 50 (86.20%), Klebsiella Pneumoni

The expression of FLI1 and its correlation with well-known breast cancer prognostic markers (ER, PR and HER2) was determined in primary breast tumors as well as four breast cancer lines including: MCF-7, T47D, MDA-MB-231 and MDA-MB-468 using RT-qPCR with either 18S rRNA or ACTB (β-actin) for normalization of data. FLI1 mRNA level was decreased in the breast cancer cell lines under study compared to the normal breast tissue; however, Jurkat cells, which were used as a positive control, showed overexpression compared to the normal breast. Regarding primary breast carcinoma

The results showed that Serum level of IL-1? was significantly increased in AD patients (3.79 ± 0.26 pg/ml) as compared with DS patients (2.78 ± 0.39 pg/ml) or controls (2.78 ± 0.22 pg/ml), while no significant difference was observed between AD and VD (3.25 ± 0.20 pg/ml) patients or between VD patients, DS patients and controls. The serum level of IL-10 was approximated in VD and DS patients and controls (3.39 ± 0.24, 2.77 ± 0.39 and 3.41 ± 0.35 pg/ml, respectively), but was significantly (P ? 0.05) increased in AD patients (5.73 ± 0.55 pg/ml
